Gastrodia Rhizome. Because there is no existing USP monograph for this dietary ingredient, a new monograph based on validated methods of analysis is proposed. The validation data for the liquid chromatographic procedure used in the test for Content of Phenols and Benzyl Citrate Glycosides were obtained using the Agilent Zorbax SB-C18 brand of column with L1 packing. The typical retention times for gastrodin, 4-hydroxybenzyl alcohol, parishin E, parishin B, parishin C, and parishin A are about 2.9, 4.6, 6.9, 8.9, 9.3, and 10.5 min, respectively. The validation data for the thin-layer chromatographic procedure used in Identification A were obtained using the Macherey-Nagel Nano-DURASIL-20/UV 254 brand of HPTLC plate; the brown band of parishin A and the dark blue band below it were well separated. When a Merck brand of HPTLC silica gel plate was used, the brown band of parishin A and the dark blue band below it were overlapped.
